چکیده

In a randomized clinical trial, study participants are assigned to various treatment arms through randomization. This random allocation of participants to treatment arms decreases bias and controls for numerous confounders (1). A common type of randomization is simple randomization, where each participant is randomly assigned to a treatment arm with a known (usually equal) probability, regardless of the treatment assignments of other study participants. For a two-arm trial, that would be achieved by tossing a fair coin. Although in the long run, on average this process will result in approximately equal numbers in each treatment arm, there is no guarantee of equally sized study groups, particularly for small samples. For example, if we want to randomize 10 participants to two treatment arms by flipping a fair coin, there is more than 6% chance of coming up with a severe imbalance as extreme as assigning eight participants to one arm and two to another arm or worse. Other randomization methods have been proposed to eliminate or at least reduce such extreme imbalance.

Randomization Time for the Overhand Shuffle

1. THE OVERHAND SHUFFLE (~o The two most common methods of shuffling a deck of playing cards are the riffle shuffle and the overhand shuffle. The riffle shuffle is performed by splitting the deck into two halves and interlacing them. Aldous and Diaconis have shown in Ref. I that approximately 2 log N riffle shuffles suffices to randomize a deck (the meaning of this will be made more precise bel...

A randomization-based perspective of analysis of variance: a test statistic robust to treatment effect heterogeneity

Fisher randomization tests for Neyman’s null hypothesis of no average treatment effects are considered in a finite population setting associated with completely randomized experiments with more than two treatments. The consequences of using the F statistic to conduct such a test are examined both theoretically and computationally, and it is argued that under treatment effect heterogeneity, use ...

Optimal Load Balancing in Publish/Subscribe Broker Networks

Load balancing in publish/subscribe (pub/sub) broker networks is challenging as the workload is multi-dimensional and content-dependent. In this paper we present the framework design of a middleware , called Shuffle, to achieve optimal load balancing in a pub/sub broker network. Shuffle features a suite of active workload management schemes within a single overlay topology on message parsing, m...

Using SAS® to Perform Robust I-Sample Analysis of Means Type Randomization Tests for Variances

A SAS macro for performing Analysis of Means (ANOM) type randomization tests for testing the equality of I variances is presented. Randomization techniques for testing statistical hypotheses can be used when parametric tests are inappropriate. Suppose that I independent samples have been collected. Randomization tests are based on shuffles or rearrangements of the (combined) sample. Putting eac...

Soluble Expression and Purification of Q59L Mutant L-asparaginase in the Presence of Chaperones in SHuffle™ T7 strain

Background and Aims: Q59L mutant of L-asparaginase enzyme from Escherichia coli (E. coli) has been introduced with lower side effects. This version of the enzyme might have potential applications in the treatment of leukemia patients. We utilized SHuffle T7 strain of E. coli, to produce the mutant enzyme in the presence of chaperone molecules. Materials and Methods: Q59LAsp gene was cloned in...
